The delivery of drugs and biologically active molecules into cells remains a major challenge in life science research. Cell-permeable peptides (CPPs), also known as cell-penetrating peptides, are short peptides that can cross the plasma membrane independently of a membrane receptor. They are typically 10-30 amino acids in length and often contain positively charged residues such as arginine and lysine.
CPPs can transport a wide range of cargo into cells, including peptides, proteins, fluorescent dyes, nanoparticles, plasmid DNA, and oligonucleotides. By attaching a therapeutic or research cargo to a CPP, intracellular delivery can often be improved without relying on viral systems. Because of their flexibility and broad utility, CPPs are widely used as non-viral transmembrane delivery vectors.
CPP-mediated delivery has shown particular value for oligonucleotide transport, but these peptides are also useful in imaging, drug delivery, cell biology, protein transduction, and targeted research applications.
