The FLAG epitope represents an 11-amino-acid leader peptide of the gene-10 product from bacteriophage T7 fused at the amino-terminus of GAL4 (yeast transcription factor).  The FLAG tag is a widely used epitope tag that can be used for the detection and purification of recombinant proteins. The LPETG motif is a recognition sequence for the transpeptidase enzyme sortase, which can be used for site-specific protein labeling and immobilization.

LPETG-flag is a 14-residue synthetic peptide with the sequence Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys-Leu-Pro-Glu-Thr-Gly-Ser. N-terminal acetylation neutralizes the terminal alpha-amino group. The sequence contains 5 Asp and 1 Glu residues, contributing anionic character near neutral pH. These sequence-derived properties describe the reagent chemically; no specific receptor, enzyme, pathway, disease association, or biological activity is assigned without product-specific experimental evidence.
