Scientific Background

Dabcyl-QALPETGEE-EDANS is an internally quenched peptide substrate for Gram-positive bacterial sortases. The central LPETG motif is a canonical Sortase A recognition sequence, while the flanking residues provide a longer sorting-signal-like context than the minimal LPETG substrate. Cleavage separates the Dabcyl quencher from EDANS and generates a fluorescence increase suitable for continuous enzyme assays.

Research Applications
  • Sortase A activity assays
  • sortase inhibitor screening
  • LPXTG substrate-recognition studies
  • bacterial transpeptidase research
Experimental Notes

The extended flanking sequence can influence substrate kinetics relative to shorter LPETG FRET constructs. Use the same substrate format when comparing inhibitor potency or enzyme variants.
