Abz-LPETG-K(Dnp)-NH2 is an internally quenched Sortase A substrate built around the LPETG recognition motif. The N-terminal anthraniloyl (Abz) fluorophore is quenched by the Dnp group before cleavage. Sortase-mediated processing of the LPETG sequence separates reporter and quencher, allowing sensitive fluorescence-based measurements of enzyme activity and substrate specificity.
