Scientific Background

DABCYL-LPETG-EDANS is an internally quenched FRET substrate containing the canonical LPXTG recognition motif of Staphylococcus aureus Sortase A. Sortase A cleaves between threonine and glycine in LPXTG-containing sorting signals and forms a transpeptidation intermediate used to anchor surface proteins to the Gram-positive cell wall. Separation of the DABCYL quencher and EDANS fluorophore upon cleavage enables continuous fluorescence-based monitoring of sortase activity.

Research Applications
  • Sortase A activity assays
  • sortase inhibitor screening
  • LPXTG recognition studies
  • transpeptidase assay development
Experimental Notes

Fluorescence can be monitored using EDANS-compatible excitation/emission settings. The substrate reports cleavage of the LPETG motif but does not reproduce every structural feature of a full-length cell-wall sorting signal.
