Scientific Background | DABCYL-LPETG-EDANS is an internally quenched FRET substrate containing the canonical LPXTG recognition motif of Staphylococcus aureus Sortase A. Sortase A cleaves between threonine and glycine in LPXTG-containing sorting signals and forms a transpeptidation intermediate used to anchor surface proteins to the Gram-positive cell wall. Separation of the DABCYL quencher and EDANS fluorophore upon cleavage enables continuous fluorescence-based monitoring of sortase activity. |
