Description:

NHS-activated Sepharose Fast Flow is a pre-activated agarose matrix for coupling amino-containing smaller proteins and peptides. NHS (N-hydroxysuccinimide) coupling forms a chemically stable amide bond with ligands containing primary amino groups. The coupled product is very stable at high pH. The product contains spacer arm between matrix and activated group. This spacer is very useful for immobilising small proteins and peptides.

Notes:
  1. Use the prepacked column for easier process.
  2. The target molecule should be stable under the selected elution conditions.
  3. Avoid air bubbles in the column.
  4. It is recommended to regenerate the resin by 4 column volumes of 0.1 M glycine-HCl (pH 2.7) after 5 separation cycles.
  5. After 10 separation cycles, wash the resin with 5 column volumes of 20% ethanol and 5~10 column volumes of 70% ethanol sequentially to remove hydrophobic substances.
  6. For longer periods of storage, keep at 4~8ºC in 20% ethanol.
