Scientific Background

(Z-Arg)2Rh110·2HCl is a bis-arginine rhodamine 110 fluorogenic substrate for arginine-directed proteases including trypsin and lysosomal cathepsins B and L. In the intact substrate, rhodamine fluorescence is strongly suppressed. Proteolytic removal of the Z-Arg groups produces highly fluorescent rhodamine 110, providing a sensitive green readout around 501 nm excitation and 527 nm emission.

Research Applications
  • trypsin activity assays
  • cathepsin B/L activity studies
  • lysosomal protease measurements
  • broad protease inhibitor screening
Experimental Notes

This is a broad arginine-directed protease substrate rather than an enzyme-specific reporter. Enzyme identity in cells or lysates should be established using selective inhibitors or orthogonal assays. Protect the rhodamine reagent from light.
