Scientific Background

(Z-Arg)2Rh110·2HCl is a rhodamine 110-based fluorogenic substrate for arginine-preferring proteases including trypsin and lysosomal cathepsins B and L. Proteolytic removal of the Z-Arg groups releases rhodamine 110 and produces a strong fluorescence increase, allowing sensitive measurement of protease activity in biochemical or cell-associated assays.

Research Applications
  • trypsin activity assays
  • cathepsin B/L activity measurements
  • flow-cytometric protease studies
  • protease inhibitor screening
Experimental Notes

The released rhodamine 110 signal is typically monitored near 501 nm excitation and 527 nm emission. This is a broad protease substrate and should not be treated as isozyme specific.
