Scientific Background | Mca-GKPILFFRLK(Dnp)-D-Arg-NH2 is an internally quenched fluorogenic substrate used for the aspartic proteases cathepsin D and cathepsin E. Cleavage within the Phe-Phe region relieves quenching of the Mca reporter and increases fluorescence. The substrate has been used in tissue and cell-extract assays to quantify cathepsin D activity and in inhibitor-screening studies. |
Research Applications | - cathepsin D activity assays
- cathepsin E activity assays
- lysosomal protease research
- aspartic protease inhibitor screening
|
Experimental Notes | Published applications commonly monitor the Mca signal near 328 nm excitation with emission in the 393-460 nm range depending on instrumentation. Pepstatin-class inhibitors can be useful as aspartic-protease controls. |
