Scientific Background

K(Biotin)-RFARKGSLRQKNV is a biotinylated form of the [Ser25]-PKC (19-31) peptide substrate. The core RFARKGSLRQKNV sequence is derived from the protein kinase C pseudosubstrate region, with the native alanine at the phosphoacceptor position replaced by serine so that the peptide can be phosphorylated. The sequence has been used extensively in PKC kinase assays, and the N-terminal biotin provides an affinity handle for streptavidin-based capture or detection formats.

Research Applications
  • protein kinase C activity assays
  • streptavidin-capture kinase assays
  • PKC inhibitor screening
  • serine phosphorylation assay development
Experimental Notes

The Ser25 substitution converts the corresponding pseudosubstrate-region sequence into a phosphorylatable substrate. The core sequence can be phosphorylated by multiple PKC isoforms and other kinases under some conditions, so it should not be described as isoform-specific.
