Scientific Background

Biotin-APRTPGGRR is an N-terminally biotinylated MAP kinase substrate based on the APRTPGGRR peptide. The PRTP segment is derived from the myelin basic protein phosphorylation region and contains the Pro-X-(Ser/Thr)-Pro motif recognized by ERK-family MAP kinases. The biotin tag provides an affinity handle for streptavidin-based capture, immobilization, and nonradioactive phosphorylation assay formats.

Research Applications
  • ERK1/ERK2 kinase assays
  • streptavidin-based phosphorylation assays
  • MAP kinase inhibitor screening
  • phosphopeptide capture and detection
Experimental Notes

Biotinylation makes this construct chemically distinct from the unlabeled APRTPGGRR substrate and can affect assay kinetics or surface interactions. Validate the labeled peptide in the intended assay format.
