What are Amyloid Beta and Amylin Peptides?

Amyloid beta (Aβ) peptides are cleavage products of amyloid precursor protein (APP) generated by β-secretase and γ-secretase processing. Among these, amyloid beta (1-42) is especially important because it is less soluble and more prone to aggregation than amyloid beta (1-40), making it a major component of amyloid plaque research.

Alzheimer’s disease is characterized by extracellular amyloid-beta deposition and intracellular neurofibrillary pathology, accompanied by progressive neuronal loss and decline in memory, cognition, and behavior. Dementia affects tens of millions of people worldwide and remains a major global health and social challenge.

Amylin, also known as islet amyloid polypeptide (IAPP), is a 37-amino acid peptide associated with amyloid formation in type 2 diabetes. Human amylin is another highly challenging peptide because it combines strong aggregation tendency with structural requirements such as a disulfide bond and C-terminal amidation.

Why Amyloid and Amylin Peptides Are Extremely Difficult to Synthesize

Amyloid peptides are among the most difficult peptide classes in solid-phase synthesis. Their production is challenging not only because of the sequence itself, but also because of their behavior during every stage of manufacturing and handling.

  • High aggregation tendency during synthesis: growing peptide chains can self-associate on resin, reducing coupling efficiency and increasing truncations.
  • Low solubility after cleavage: many amyloid sequences are difficult to dissolve reproducibly, especially at higher concentrations.
  • Hydrophobic sequence regions: these regions complicate chain elongation, purification, and analytical characterization.
  • Rapid self-assembly in solution: oligomerization and fibril formation can begin during handling, affecting reproducibility.
  • Amylin-specific structural requirements: human amylin requires correct disulfide bond formation and C-terminal amidation in addition to aggregation control.
