Scientific Background

H-D-Val-Leu-Lys-AFC is a short fluorogenic peptide substrate used for plasmin activity measurements. Proteolytic cleavage at the lysine-AFC bond releases 7-amino-4-trifluoromethylcoumarin (AFC), producing a fluorescence signal suitable for continuous kinetic assays. The substrate has been used in published plasmin and plasminogen-activation studies, including measurements in plasma and bacterial-fibrinolysis models.

Research Applications
  • plasmin activity assays
  • fibrinolysis research
  • plasminogen-activation studies
  • serine-protease inhibitor screening
Experimental Notes

AFC is commonly monitored near 380 nm excitation and 500 nm emission. Although widely used as a plasmin readout, activity in complex samples should be interpreted with appropriate protease and inhibitor controls.
