Mca-PLGLEEA-Dap(Dnp)-NH2 is an internally quenched fluorogenic substrate developed for matrix metalloproteinase-12 (macrophage elastase). Proteolytic cleavage separates the Mca fluorophore from the Dnp quencher and increases fluorescence, enabling continuous MMP-12 activity measurements. The substrate was developed in studies aimed at improving selectivity for MMP-12 over related metalloproteinases.
