Scientific Background

Z-Phe-Arg-AMC is a fluorogenic peptide substrate for kallikrein and related arginine-directed serine proteases. Proteolytic cleavage at the Arg-AMC bond releases 7-amino-4-methylcoumarin, producing a strong fluorescence increase that enables continuous activity measurements. The Z-Phe-Arg recognition motif has been used in classical comparisons of kallikrein, thrombin, factor Xa and urokinase substrate preferences.

Experimental Notes

Arg-AMC substrates can be cleaved by more than one trypsin-like serine protease. Use purified enzyme, selective inhibitors or orthogonal controls when assigning activity in complex samples. Protect AMC-containing substrate from light.
