This peptide contains trimethylated Lys12 within the H4 N-terminal tail, together with an N-terminal acetyl group and C-terminal biotin handle. It provides a defined methyl-lysine reagent for studying uncommon H4 methylation states and modification-sensitive binding.

H4K12me3 is distinct from the much more common H4K12ac mark. The trimethylated lysine state should be preserved exactly when evaluating reader or antibody specificity.
