Scientific Background

Ac-DE-Dap(QXL520)-EE-Abu-ψ-[COO]AS-C(5-FAMsp)-NH2 is a highly sensitive internally quenched FRET substrate for hepatitis C virus NS3/4A protease. The sequence contains an engineered scissile ester linkage within an HCV protease recognition motif, together with a QXL 520 quencher and 5-FAM reporter. Cleavage by NS3/4A separates the reporter pair and produces a fluorescence increase suitable for continuous protease activity measurements.
