Scientific Background | 6-FAM-VFDAVTDVIIKNNLKECGLY is a fluorescein-labeled 20-residue peptide derived from the C-terminal region of the Gαi3 protein and is commonly referred to as HCAM-2 or F-Gαi3C20. Pertussis toxin catalyzes ADP-ribosylation of a cysteine near the C-terminus of inhibitory G-protein alpha subunits. This synthetic peptide preserves the relevant C-terminal cysteine-containing recognition region and has been used as a defined substrate in in vitro assays that measure the ADP-ribosyltransferase activity of pertussis toxin. |

Experimental Notes | The 6-FAM label supports fluorescence-based detection, but pertussis-toxin modification is an ADP-ribosylation reaction rather than proteolytic cleavage. Published assay formats commonly separate and quantify the ADP-ribosylated peptide product by HPLC. The peptide should therefore not be described as a conventional fluorogenic protease substrate. |
