Scientific Background | 5-FAM-KKKKEEIYFFFG-NH2 is a fluorescent tyrosine-containing peptide substrate commonly called FAM-CSKtide. The unlabeled KKKKEEIYFFFG motif has been used to measure C-terminal Src kinase (Csk) activity and is also compatible with a broad panel of receptor and non-receptor tyrosine kinases. The N-terminal 5-FAM reporter supports fluorescence-based detection and separation workflows while the Tyr residue serves as the phosphorylation site. |
Research Applications | - Csk kinase activity assays
- receptor tyrosine kinase profiling
- fluorescence-based phosphorylation assays
- kinase inhibitor screening
|
Experimental Notes | CSKtide is not exclusive to Csk and can be phosphorylated by several tyrosine kinases. The 5-FAM label also changes mass, charge and chromatography relative to the unlabeled peptide, so quantitative assays should use the labeled reagent itself for calibration. |
