Scientific Background

(Z-Ala-Arg)2Rh110·2HCl is a rhodamine 110-based fluorogenic substrate for elastase, trypsin and related serine-protease activity measurements. Cleavage of the peptide groups releases fluorescent rhodamine 110, producing a strong signal suitable for continuous or endpoint assays. Rhodamine-based dipeptide substrates were developed as highly sensitive reporters for serine proteinases.

Experimental Notes

The released Rh110 signal is monitored near 497 nm excitation and 527 nm emission. This is not an isozyme-specific substrate, so purified-enzyme or inhibitor controls are recommended when assigning protease identity.
