Scientific Background

Suc-LLVY-AMC is a fluorogenic tetrapeptide substrate used for chymotrypsin-like protease activity measurements, including calpains and the 20S proteasome. Proteolytic cleavage releases AMC and generates a fluorescence signal that can be monitored continuously. Because multiple protease systems can hydrolyze LLVY-AMC substrates, the reagent is best used with selective inhibitors or purified enzymes when assigning activity to a specific protease.

Research Applications
  • calpain activity assays
  • 20S proteasome activity studies
  • chymotrypsin-like protease assays
  • protease inhibitor screening
Experimental Notes

AMC is typically monitored near 353 nm excitation and 442 nm emission. Protect the substrate from light and use pathway-selective inhibitors when measuring activity in complex lysates.
