Autocamtide-2 (AC-2), KKALRRQETVDAL, is a well-established peptide substrate for Ca2+/calmodulin-dependent protein kinase II (CaMKII). It is derived from the CaMKII autophosphorylation region and contains a threonine phosphoacceptor in a recognition sequence efficiently phosphorylated by activated CaMKII. AC-2 is frequently used as a positive control and quantitative substrate in CaMKII activity assays.
