Scientific Background | Mca-Ala-Pro-Lys(Dnp) is an internally quenched fluorogenic substrate widely used to measure angiotensin-converting enzyme 2 (ACE2) catalytic activity. The Mca fluorophore is quenched by the Dnp group before cleavage; ACE2-mediated hydrolysis separates the reporter and quencher and produces a large fluorescence increase. This reagent was used in biochemical characterization of ACE2 peptide-hydrolysis specificity. |
Research Applications | - ACE2 activity assays
- renin-angiotensin system research
- ACE2 inhibitor screening
- FRET peptidase kinetics
|
Experimental Notes | ACE2 activity assays require control of other metallopeptidases and peptidases that may process short fluorogenic substrates. Appropriate inhibitor cocktails and ACE2-selective controls are recommended for complex samples. |
