FAM-QTNSPRRARSVASQSK-QXL520-NH2 is an extended internally quenched substrate centered on the SARS-CoV-2 spike S1/S2 polybasic cleavage region. The sequence includes the QTNSPRRARSVASQS context surrounding the RRAR motif and places FAM and QXL520 at opposite ends of the reporter construct. Cleavage produces a fluorescence increase suitable for protease-activity and inhibitor-screening studies.
