Scientific Background

Pyrrhocoricin is a 20-residue proline-rich antimicrobial peptide originally isolated from the firebug Pyrrhocoris apterus. Unlike strongly membrane-lytic antimicrobial peptides, pyrrhocoricin has been shown to act intracellularly in susceptible bacteria by binding the DnaK/Hsp70 chaperone system and interfering with protein folding. Its proline-rich sequence has also been used as a scaffold for structure-activity studies aimed at improving antibacterial potency and stability.

Research Applications
  • proline-rich antimicrobial peptide research
  • DnaK/chaperone interaction studies
  • antibacterial structure-activity studies
  • microbial growth-inhibition assays
Experimental Notes

Pyrrhocoricin activity is species dependent and should not be generalized across all bacteria. Assay medium, ionic strength, inoculum and peptide concentration can materially affect measured antibacterial activity.
