Scientific Background

Myristoylated AC3-I is an inhibitory peptide derived from Autocamtide-3 and designed to inhibit Ca2+/calmodulin-dependent protein kinase II (CaMKII). Replacement of the substrate phosphoacceptor residue with alanine creates a nonphosphorylatable inhibitory sequence, while N-terminal myristoylation increases hydrophobicity and facilitates use in cellular signaling studies. AC3-I has been used to examine the contribution of persistent CaMKII activity to neuronal plasticity.

Research Applications
  • CaMKII inhibition studies
  • synaptic plasticity research
  • Ca2+/calmodulin signaling
  • cell-associated kinase inhibitor experiments
Experimental Notes

This reagent is an inhibitor rather than a phosphorylation substrate. Myristoylation affects membrane partitioning and solubility, and experimental conclusions should use sequence-appropriate inhibitor controls.
