Scientific Background | This internally quenched Mca/Dnp peptide is a fluorogenic matrix metalloproteinase substrate. A closely corresponding Mca-Pro-Cha-Gly-Nva-His-Ala-Dnp-containing substrate was used in biochemical characterization of human collagenase-3 (MMP-13), where it was efficiently hydrolyzed. The substrate format is also responsive to MMP-8 and MMP-9, making it useful for comparative metalloproteinase activity measurements rather than as an absolutely isoform-specific reagent. |
Research Applications | - MMP-8, MMP-9 and MMP-13 activity assays
- FRET-based metalloproteinase measurements
- MMP inhibitor screening
- TIMP inhibition studies
|
Experimental Notes | Mca fluorescence is quenched by the Dnp group until proteolytic cleavage separates the reporter and quencher. Enzyme specificity depends on assay conditions and should be confirmed experimentally. |
