Scientific Background

This internally quenched Mca/Dnp peptide is a fluorogenic matrix metalloproteinase substrate. A closely corresponding Mca-Pro-Cha-Gly-Nva-His-Ala-Dnp-containing substrate was used in biochemical characterization of human collagenase-3 (MMP-13), where it was efficiently hydrolyzed. The substrate format is also responsive to MMP-8 and MMP-9, making it useful for comparative metalloproteinase activity measurements rather than as an absolutely isoform-specific reagent.

Experimental Notes

Mca fluorescence is quenched by the Dnp group until proteolytic cleavage separates the reporter and quencher. Enzyme specificity depends on assay conditions and should be confirmed experimentally.
