Scientific Background

DABCYL-GABA-SQNYPIVQ-EDANS is an internally quenched fluorogenic substrate for HIV-1 protease. The peptide is derived from a natural retroviral processing-site sequence and is cleaved at the Tyr-Pro bond. Proteolysis separates DABCYL from EDANS, producing a fluorescence increase that enables continuous kinetic measurements of HIV-1 protease activity and inhibitor effects.

Research Applications
  • HIV-1 protease activity assays
  • retroviral protease kinetics
  • HIV protease inhibitor screening
  • continuous FRET protease assays
Experimental Notes

The substrate is commonly dissolved initially in concentrated DMSO because of limited aqueous solubility. Protect the fluorescent substrate from light and avoid repeated freeze-thaw cycles after reconstitution.
