Scientific Background | QXL520-GABA-SQNYPIVQ-K(HiLyte Fluor 488)-NH2 is a long-wavelength FRET substrate for HIV-1 protease. The SQNYPIVQ sequence derives from the p17/p24 cleavage junction of the HIV Gag precursor, an essential viral maturation site. The HiLyte Fluor 488/QXL520 pair shifts detection to approximately 490/520 nm, reducing short-wavelength autofluorescence and providing a sensitive continuous readout of HIV protease activity. |
Research Applications | - HIV-1 protease activity assays
- antiretroviral protease inhibitor screening
- viral polyprotein-processing research
- continuous FRET protease measurements
|
Experimental Notes | This reporter construct is chemically distinct from EDANS/DABCYL HIV protease substrates even though the underlying cleavage motif is related. Kinetic and inhibitor data should be compared using matched substrate chemistry. |
