HDAC Fluorogenic Substrate, Ac-RGK(Ac)-AMC

Ac-RGK(Ac)-AMC is a fluorogenic histone deacetylase substrate containing an epsilon-acetylated lysine adjacent to an AMC reporter group. In the original assay design, HDAC-catalyzed deacetylation is followed by a developer step that releases fluorescent AMC, providing a sensitive format for measuring HDAC activity and screening deacetylase inhibitors.

This substrate generally requires deacetylation followed by a developer/protease step to liberate AMC. It should not be interpreted as a simple one-step protease substrate.
