5-FAM-YGRKKRRQRRRCSTRIRRQL-NH2 is a fluorescently labeled form of gp91ds-tat, a cell-permeable peptide developed to interfere with assembly of the phagocyte NADPH oxidase complex. The parent gp91ds-tat combines an HIV Tat-derived cell-penetrating sequence with a gp91phox/NOX2-derived sequence that interacts with p47phox. The 5-FAM modification provides a fluorescent handle for localization, uptake and assay-development studies involving the gp91ds-tat reagent.

Experimental Notes

The fluorescent derivative should not automatically be assumed to have identical inhibitory potency to unlabeled gp91ds-tat. The parent peptide is also not absolutely NOX2-specific because homologous oxidase sequences can contribute to activity in some systems.
