CK2 Substrate Peptide, RRRDDDSDDD

RRRDDDSDDD is an acidic synthetic substrate for protein kinase CK2. The peptide contains the characteristic acidic environment favored by CK2 around the phosphoacceptor serine and has been used in kinetic studies of CK2 holoenzyme and catalytic subunits. Published work reported an apparent Km of approximately 60 micromolar for CK2 under 150 mM NaCl conditions, illustrating the utility of this defined peptide for quantitative kinase assays.

CK2 peptide kinetics can depend strongly on ionic strength and enzyme subunit composition. Buffer composition should therefore be controlled when comparing kinetic parameters.
