Scientific Background | FRETS-VWF73 is a long internally quenched peptide substrate derived from the von Willebrand factor A2 domain and designed for sensitive measurement of ADAMTS13 activity. The construct incorporates an N-methylanthranilic acid fluorophore on Dap and a Dnp quencher around the physiologic Tyr-Met cleavage region. Proteolysis by ADAMTS13 separates the reporter pair, enabling rapid fluorescence-based determination of VWF-cleaving protease activity. |
Research Applications | - ADAMTS13 activity assays
- von Willebrand factor proteolysis research
- thrombotic thrombocytopenic purpura studies
- plasma ADAMTS13 assay development
|
Experimental Notes | ADAMTS13 cleavage of VWF is strongly influenced by substrate conformation. FRETS-VWF73 is an engineered soluble assay substrate rather than intact multimeric VWF, so kinetic results should be interpreted within the assay format. |
