Scientific Background

[D-Trp12, Tyr34]-pTH (7-34) amide (bovine) is a synthetic research peptide in the parathyroid hormone / PTH-related peptide family. Parathyroid hormone (PTH) and PTH-related peptide (PTHrP) signal through PTH1R, a class B GPCR. Structural and structure-activity studies support a two-part interaction model in which the C-terminal portion of PTH(1-34) contributes strongly to receptor binding while the N-terminal portion is important for receptor activation. PTH/PTHrP fragments, substitutions, and terminally modified analogs are therefore useful for dissecting binding, activation, and peptide conformation.

Research Applications
  • receptor or ligand-binding assays
  • competition studies
  • biochemical or enzymatic assays
  • analytical LC-MS/HPLC methods
  • antibody controls
  • cell-based experiments
  • comparative structure-activity studies
Experimental Notes

For a fragment, residue-substituted analog, stereochemical variant, labeled peptide, or terminally modified form, experimental behavior should be evaluated for the exact construct rather than assumed to match the native parent peptide. Sequence-dependent solubility, aggregation, adsorption, oxidation, and stability should be considered when establishing reconstitution, storage, and assay conditions.
