Product Description

Cy5-TAT (48-60) Cell-Penetrating Peptide contains the classic HIV-1 Tat-derived cell-penetrating peptide sequence GRKKRRQRRRPPQQ. Tat peptides are among the best-known arginine-rich protein transduction domains and are widely used as model CPPs in studies of intracellular delivery.

Tat(48-60) Cell-Penetrating Peptide

The Tat basic region is enriched in arginine and lysine residues, giving the peptide a strong positive charge under physiological conditions. This cationic character promotes interaction with anionic components of cell surfaces, including glycosaminoglycans, and contributes to uptake through endocytic and membrane-associated mechanisms.

Tat-derived CPPs have been investigated for delivery of peptides, proteins, oligonucleotides, nanoparticles, and other research cargos. Uptake mechanism and intracellular localization can depend strongly on cargo, concentration, cell type, and experimental conditions.

Cy5-Labeled Configuration

LT8678 adds a cysteine that serves as a site-selective attachment point for Cy5. The far-red fluorophore allows direct measurement of peptide binding and uptake while leaving the published Tat sequence intact.

  • CPP uptake studies
  • Flow cytometry
  • Live- or fixed-cell fluorescence imaging
  • Peptide trafficking studies
  • Cargo-delivery comparisons
  • Membrane interaction research

Published Tat Sequence

GRKKRRQRRRPPQQ

The cysteine/Cy5 functionality in LT8678 is an added analytical label and not part of the canonical Tat sequence.
