Scientific Background

Caspase 1 Substrate 2m (ICE), fluorogenic; Ac-YVAD-AMC is a synthetic research peptide in the caspase peptide substrate family. Caspases are cysteine proteases that recognize short Asp-containing peptide motifs. Positional-scanning and structural studies established distinct substrate preferences among caspase family members; DEVD-type sequences are widely used for caspase-3/7 activity, whereas YVAD-related motifs are associated with caspase-1 assays. AMC- and pNA-labeled tetrapeptides permit fluorogenic or chromogenic detection of peptide cleavage.

Research Applications
  • caspase activity assays
  • apoptosis-related protease measurements
  • fluorogenic AMC assays
  • chromogenic pNA assays
  • substrate-specificity studies
Experimental Notes

The exact sequence, reporter group, phosphorylation state, stereochemistry, and terminal modifications should be considered when comparing this reagent with the parent peptide or published assay substrates. Enzyme concentration, buffer composition, cofactors, pH, ionic strength, and substrate concentration can also affect measured activity.
