Description:

Recharging Ni-NTA Resin

Ni-NTA resin can be recharged following the procedures below:

- For 1 ml of resin, wash with 2 ml of 7 M GuHCl.

- Wash resin with 5 ml of deionized water.

- Wash resin with 3 ml of 2% SDS.

- Wash resin with 5 ml of deionized water.

- Wash resin with 5 ml of 100% EtOH

- Wash resin with 5 ml of deionized water.

- Wash resin with 5 ml of 100 mM EDTA (pH 8) to strip Ni+2 ions.

- Wash resin with 5 ml of deionized water.

- Wash resin with 5 ml of 100 mM NiSO4.

- Wash resin with 10 ml of deionized water.

- Preserve resin in 0.02% azide or 20% ethanol.

       Recombinant Staphylococcal Protein A Agarose

Notes:

  1. Avoid air bubbles.
  2. Regenerate the resin every 5 purification in order to maintain the product efficiency.
  3. After every 10 separation cycle, wash the resin with 5 column volumes of 20% ethanol and 5~10 column volumes of 70% ethanol sequentially to remove hydrophobic substances.
  4. Keep at 4~8 °C in 20% ethanol.
